Immune Cell Killing and Clearance Assays

Immune Cell Killing and Clearance Assays

Immune-cell recognition, killing and clearance of diseased cells and pathogens are critical components of the human host defense system. The IncuCyte live-cell analysis system enables real-time visualization and measurements of T cell killing and the phagocytic clearance of pathogens or dying cells by phagocytes.

IncuCyte® Immune Cell Killing Assays

  • Visualize and automatically analyze the entire time course of immune cell killing using IncuCyte® apoptosis reagents and IncuCyte® live-cell labeling reagents.

video t-cell killing

Real-time monitoring of immune cell killing. IncuCyte movies showing a T cell sub population of PBMCs, activated with anti-CD3 antibody and IL-2, killing target SK-OV-3 tumor cells. Labeling of the target cells with IncuCyte® NucLight Red Live-Cell Labeling reagent enables direct counting of viable target cell numbers over-time. Addition of the IncuCyte® Caspase 3/7 apoptosis reagent enables simultaneous detection of cells undergoing apoptosis (green fluorescence).

IncuCyte® Phagocytosis Assays

Phagocytosis of Cells

  • Automatically measure phagocytosis of diseased, dead or dying target cells
  • Investigate antibody mediated cellular phagocytosis (e.g., anti-CD47 antibody mediated phagocytosis / ADCP) or efferocytosis (phagocytosis of apoptotic cells).

video phagocytosis of cells

Real-time monitoring of phagocytosis of apoptotic cells. IncuCyte® pHrodo® Red Labeled apoptotic Jurkat T lymphocytes engulfed by the mouse macrophage cell type J774A.1. Apoptotic Jurkat cells are phagocytosed and, on entering the acidic environment of the phagosome, increase in fluorescence.

Phagocytosis of Bioparticles®

  • Real-time visualization and analysis of the internalization of Bioparticles® using pH sensitive-conjugated probes
  • Monitor the phagocytosis of bacterial Gram positive, Gram negative or yeast-derived pathogens

video phagocytosis of bioparticles

Real-time monitoring of phagocytosis. IncuCyte® pHrodo® Green E. coli Bioparticles® engulfed by the mouse macrophage cell type J774A.1. The Bioparticles® are phagocytosed and, on entering the acidic environment of the phagosome, increase in fluorescence. Note the dynamic activity of the macrophages and engulfment of bioparticles over time via the formation of phagocytic cups.
