Immune Cell Killing, Single Spheroid

Watch how Herceptin alone, or peripheral blood mononuclear cells (PBMCs) alone, have no significant effect on the growth of a SKOV-3 ovarian tumor spheroid. Whereas a combination of Herceptin and PBMCs so effectively kill a SKOV-3 spheroid. These stunning phase and fluorescence images were collected continuously without interruption, over a 10 day interval.
It is fascinating to watch antibody-dependent cell-mediated cytotoxicity (ADCC) in a 3D cell culture model, being monitored. HER2-positive SKOV-3 spheroids formed (3 days) in 96-well ULA plates were treated with Herceptin (mAb targeting HER2 receptors) and PBMCs (E:T, 2.5:1).
