Apoptosis

Apoptosis

ApoptosisKinetic induction of NF-kB driven rhGFP reporter gene expression following rhTNF-alpha stimulation in HEK293 cells (n=5 wells).

Apoptosis

96-well IncuCyte Platemap showing 5 replicate wells for decreasing concentrations of TNF-alpha (Col 4-11). Not all wells were used in this experiment. Columns 2 and 3 were control wells. Vertical axis is fluorescent object confluence, horizontal axis is time.

KEY FEATURES

  • Data rich: 96-well real-time kinetic data offering insight not attainable with end-point assays
  • Cost effective: does not require a terminal reaction substrate as with luciferase assays saving time and money
  • Convenient: kinetic read-out enables the user to optimize the signal window within one experiment, without having to make decisions a priori as to when to stop the experiment
  • Sensitive: provides multiple temporal data points per condition, enhancing the quantitative robustness of the assay

ASSAY EXAMPLE

NF-kB Reporter Assay on IncuCyte:   In this example, we used a commercially available reporter plasmid to transiently transfect HEK cells. We then used the IncuCyte FLR to automatically acquire images at 3 hour updates and the IncuCye analysis software to quantify temporal NF-kB transcription resulting from TNFalpha receptor activation. Running a reporter assay in this fashion has many advantages over traditional luciferase assays as described above.

Reporter Assay Services: Customers also have the option of using our Assay Services for assay development or drug screening. In this model, customers can take advantage of our experience in running reporter assays to quickly answer either proof-of-concept or drug screening questions. This frees up internal resources if outsourcing is preferred or provides an entry point for evaluating IncuCyte technology prior to making a purchase.
